Resumen : El propósito de esta investigación fue identificar morfolögicamente el agente causal de la mazorca negra en cacao en los cantones de Quinindé y Calceta, como también evaluar el comportamiento patogénico, cultural y sensibilidad in vitro de aislados de Phytophthora spp provenientes de los cantones de Quinindé y Calceta. Esta investigación se ejecutó en los laboratorios de Biología Molecular de la ESPAM – MFL, Calceta – Ecuador. Se colectaron frutos sintomáticos en Calceta y Quinindé respectivamente, para la identificación del patógeno se aislaron las cepas y se las observaron en el microscopio, donde se visualizó las estructuras principales del patógeno como son esporangios y clamidosporas unidas al micelio, se realizó la patogenicidad y agresividad de las cepas obtenidas donde se corroboró posteriormente a los 2 y 3 días de la inoculación, los aislados de Quinindé fueron mas agresivos frente a los de la Calceta, los fungicidas (Carboxin-Thiram, Mancozeb-Cymoxanil, Mancozeb-Metaxyl y Metalaxyl), empleados en la prueba de sensibilidad in vitro controlaron el crecimiento del patógeno, en la prueba de caracterización cultural las cepas con mayor crecimientos fueron las sembradas en medio de cultivo Agar Zanahoria para ambos cantones, los patrones culturales que se presentaron en el ensayo de caracterización cultural fueron aracnoide en los medios EMA, PDA, SDA y estrellado en AZ.
Descripción : The purpose of this investigation was to morphologically identify the causal agent of black pod in cocoa in the cantons of Quinindé and Calceta, as well as to evaluate the pathogenic, cultural and in vitro sensitivity behavior of isolates of Phytophthora spp from the cantons of Quinindé and Calceta. This research was carried out in the Molecular Biology laboratories at ESPAM - MFL, Calceta - Ecuador. Symptomatic fruits were collected in Calceta and Quinindé respectively, for the identification of the pathogen the strains were isolated and observed under the microscope, where the main structures of the pathogen such as sporangia and chlamydospores attached to the mycelium were visualized, pathogenicity and aggressiveness were performed of the strains obtained where it was corroborated 2 and 3 days after inoculation, the Quinindé isolates were more aggressive compared to the Calceta isolates, the fungicides (Carboxin-Thiram, Mancozeb-Cymoxanil, Mancozeb-Metaxyl and Metalaxyl), employed in the in vitro sensitivity test controlled the growth of the pathogen, in the cultural characterization test the strains with the highest growth were those sown in Agar Carrot culture medium for both cantons, the cultural patterns that were presented in the cultural characterization test They were arachnoid in the EMA, PDA, SDA media and crashed in AZ.
